Would the .forward file need to be associated with a login/password?

There would be 500+ forwarding accounts - I'm not sure if for this virtual domain, we need to setup 500 logins/password, or if we can simply link it to flatfile/database and somehow amend forwarding for say, user234@xxxxxxxxxxxxxxx without having a login.
