Yes. If you give a user a 10MB quota, they can store up to 10MB of any 
files with their username as the "owner"... that includes the mail 
spool, files in their home dir, or files anywhere else on the disk 
that they ended up with write permission to and stashed stuff (/tmp, 
etc).
