Yes.

That's how SquirrelMail works by default. Follow the instructions in the
INSTALL file of the SquirrelMail distro.

Yes that all works. SquirrelMail uses IMAP folders so if your users move
mail from their mail spool (inbox) to a folder those messages won't be
available via POP though.
