Enter the following values in the virtual site setup sceen (using your example):

Host name - sub
Domain name - domain.co.uk
Web Server Alias - leave blank
Mail Server Alias - leave blank

giving you http://sub.domain.co.uk ...

That's all there is to it.  Here is an example of such a sub-domain site that we
host:  http://dine.waterfront.co.za/
